QUÍMICA D’ÀCIDS NUCLEICS

La síntesi d’àcids nucleics està en constant evolució i per això ens pot proporcionar un gran nombre d’eines molt versàtils amb diverses aplicacions tant en teràpia com en ciència dels materials. Els nostres estudis estan orientats a la millora del coneixement dels àcids nucleics mitjançant de la introducció de modificacions amb l’objectiu d’avaluar la seva eficàcia en teràpies tals com la teràpia antisentit i l’ARN d’interferència. Els projectes portats a terme pel nostre grup estan basats en estudis estructurals d’àcids nucleics amb especial èmfasi en aquells amb nucleòtids modificats o conjugats. A més a més, s’està treballant en nous agents de transfecció basats en els lípids catiònics per millorar l’administració dels àcids nucleics. Finalment, ressaltar el desenvolupament de noves molècules d’àcids nucleics per l’assemblatge de nanomaterials i la utilització de nous sistemes biosensors per detectar ADN i ARN biològicament rellevants.

- Tríplex: modificacions químiques i estudis d’estabilitat estructural.
- Quàdruplex de guanina: modificacions químiques en els “loops” i les tètrades, noves arquitectures, disseny de lligands específics per seqüencies de rellevància biològica.
- “i-Motif”: caracterització fisico-química, disseny de lligands específics per seqüencies de rellevància biològica, avaluació quantitativa dels equilibris dúplex / “i-motif”
- Conjugats dels àcids nucleics: lípids, carbohidrats, agents intercalants, pèptids/PNA, colorants, polímers
- Oligonucleòtids que contenen bases non-naturals de rellevància estructural i biològica
Oligonucleòtids terapèutics
- RNA interferents, siRNA: millora de l’estabilitat, eficàcia de inhibició de la expressió gènica i selectivitat (reducció d’efectes no desitjats); caracterització estructural de les interaccions siRNA-proteïna; sistemes d’administració.
- ADN Antisense: modulació de la estabilitat a les nucleases; desenvolupament de sistemes d’administració utilitzant nous agents de transfecció.
- microRNA: miRNA mimètics i inhibidors (antagomirs)
- Aptàmers: increment del temps de vida mitja; transportadors de drogues citotòxiques.
Nanotecnologia de l’ADN (aplicacions biomèdiques i biosensors)
- Preparació d’estructures a escala nanomètrica dirigides por l’ADN (papiroflèxia de l’ADN o ADN “origami”, ADN tetraèdric).
- Nanomotlles d’ADN: Biosensors para la detecció de trombina i d’enzims de reparació de l’ADN; sistemes d’administració de drogues citotóxiques, motlles moleculars per al litografiat de superfícies d’or.
- Sondes basades en l’ADN para a la detecció de miRNA i de la metilació de l’ADN.
